Is it possible to unlock a locked layout in MFME, considering it has been published?

No!! Now why would you want to do that I ask
What layout are you trying to hack?
Ask the original creator to see if they can lend you an unlocked WIP layout?
Is it for another rehash layout or something more sinister??

If it's one of yours, look for the following file, in the layout folder.

 

(the layouts name)_unpub.fml and rename it to (the layouts name).fml file and overwrite it.

 

If it's not and the layout producer, hasn't left the unpub.fml file in the release folder, than you can't without getting it from the layout producer themself.

Was pissing about. From what I have analysed I doubt that but we have already been there and that subject is old hat besides, there is no point dwelling over the past and shouldn't of joked about it really.
