to use with mess, you will need to rename the mame.ini to the name of the system and drop it in the ini folder in mess, so genesis.ini , delete , rename mame.ini to genesis.ini and your good to go

hmm dunno, ive remade the config file

Will work with any monitor/hd tv/whatever else you've rigged up as a display
removed invalid paths
copy into mame\ini and overwrite the mame.ini in there , if your not happy, delete the file and mame will recreate it
dunno why you dont have any permission to use, if its mame, check you have admin access or right click mame.exe and run as administrator, as for dropbox, try this mediafire link instead, if people get really stuck i will upload my custom mame into a zip and put the whole lot in there.

to use that mame.ini , copy in to mame/ini directory, go into OPTIONS / Directories and point it to your mame roms folder, i just tried my config with mess 0.154 and mameuifx 0.154 , lol i know its annoying rescanning your roms ( if you have a fullset ) if your running on a default config and want all the games / stuff to look like its being emulated on a CRT/oldskool monitor this config is close to arcade perfect, copy out your original mame config ( mame/ini/mame.ini and replace it with the one in the link above, does put the games into original output size and adds tiny blur, proper colour emulation and a few other bits and bobs,

yeah i need to debug the input , double check the match and throw it onto 48, thanks for the info, i need to break down the 2.1 source and have a good read through. Im still working on mame emu stuff, have just finsihed some HLSL settings that make the video output look pretty much arcade perfect. Going to post that for all fo you now......
